Senthil and Ganga marry against the wishes of their extended family, and move into a spectacular mansion in the same village, despite fears of it being haunted. Soon after moving into their new abode, strange things begin to happen; whether these are caused by the apparent ghost of a former occupant, Chandramukhi, or any one of the suspicious members of the extended family, is investigated by the psychiatrist extraordinaire, Dr Saravanan. Will he be successful in unraveling the mystery and protect the family from harm?

A sequel to "Chandramukhi", tentatively titled "Chandramukhi 2" in Tamil, is currently being made by P Vasu, who directed the sequel to "Apthamitra", titled "Aptharakshaka".

On 29 February 2008, producer AM Rathnam dubbed and released the film in Hindi, despite "Bhool Bhulaiyaa", the Bollywood remake of "Manichithrathazhu", having been released on 12 October 2007. The film was also dubbed in Bhojpuri with the title "Chandramukhi Ke Hunkaar".

After the phenomenal success of the movie, it was dubbed into Telugu with the same title, "Rajmohol" (2005) in Bengali. Later, it became the first Tamil film to be dubbed into German and it was released in several German-speaking countries as "Der Geisterjäger" (lit. The Ghost Hunters).

After the audio launch, Rajinikanth and Nayantara shot a song in Turkey, which was the first ever time in the history of Tamil cinema; while, the song "Athinthom" was shot at a Mysore palace.

Lead actress, Jyothika won many awards for her excellent performance in the title including a Tamil Nadu State Film Award for Best Actress, a nomination for Filmfare Best Actress among several other awards.

The film was awarded Tamil Nadu State Film Award for Best Film for year 2005.

Rajnikanth saw the movie in Bengaluru and was very much impressed with the way his long time friend Vishnuvardhan had portrayed the role. He decided to play the character in the Tamil version of the movie, and made "Chandramukhi" his "come back" film after the box office failure of "Baba" (2002).

It's a remake of the Kannada film "Apthamitra", which in itself was a remake of the Malayalam film "Manichitrathazhu".
